Product Specific Information

The monoclonal antibody bg1fpj recognizes mouse CD369 (Dectin-1, CLEC7A), a type II transmembrane protein in the C-type lectin family. Its ligand is β1,3-glucan, a key polysaccharide component in the cell wall of fungi, plants, and some bacteria.
CD369 binds organisms such as Candida, Aspergillus, Pneumocystis, Coccidioides, and Mycobacterium. Upon ligand binding, calcium-dependent signaling via the ITAM domain activates Syk kinase, leading to endocytosis/phagocytosis and cytokine production (IL-2, IL-6, IL-10, TNF-α, IL-1β, IL-23).
CD369 signaling contributes to Th17 and Th1 differentiation and is mainly expressed on myeloid cells (neutrophils, monocytes, macrophages, dendritic cells), with lower expression on lymphocytes and mucosal epithelium.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of mouse lysed whole blood. Recommended ≤0.125 µg/test (100 µL final volume). Optimize empirically per assay (10⁵–10⁸ cells/test).
Light Sensitivity: Protect from light; tandem dye is sensitive to photo-induced oxidation.
Fixation: Samples can be stored up to 3 days at 4°C in the dark using:

  • IC Fixation Buffer (Product #00-8222-49)
  • 1-step Fix/Lyse Solution (Product #00-5333-54)
    Fluorophore performance may vary by clone; confirm empirically.

Target Information

CLEC7A (Dectin-1) is a member of the C-type lectin-like receptor family, predominantly expressed on myeloid cells. It is a small type II membrane glycoprotein with an extracellular C-type lectin-like domain and a cytoplasmic ITAM motif.
CLEC7A functions as a pattern-recognition receptor that detects β-1,3- and β-1,6-linked glucans from fungi and plants, triggering innate immune responses through Syk kinase activation and reactive oxygen species production.
