Product Specific Information

The FJK-16s antibody reacts with mouse, rat, dog, porcine, bovine, and cat Foxp3 (FORKHEAD BOX P3, SCURFIN, JM2). Cross-reactivity with other proteins has not been determined. Foxp3 (49–55 kDa) is a transcriptional regulator associated with regulatory T cells (Treg). The FJK-16s antibody epitope maps to amino acids 75–125 of mouse Foxp3. In humans, this region is alternatively spliced, with isoforms present in CD4+CD25+ lymphocytes.

FJK-16s is optimized for use with the Foxp3/Transcription Factor Buffer Staining Set (Product #00-5523-00). Use of other fixation or staining buffers is not recommended.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Intranuclear staining and flow cytometry using the Foxp3/Transcription Factor Buffer Set. Recommended ≤0.5 µg/test (100 µL volume, 10⁵–10⁸ cells/test). Optimize antibody titration for best performance.

Light Sensitivity: Protect from light; tandem dye is photo-sensitive.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark.
Excitation/Emission: 488–561 nm / 775 nm; compatible with Blue, Green, and Yellow-Green lasers.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

FOXP3 (Forkhead box protein 3) is a transcriptional regulator essential for immune homeostasis. It is highly expressed in CD25+CD4+ regulatory T cells and absent in CD8+ T cells. FOXP3 defects cause the scurfy phenotype in mice and IPEX syndrome (immune dysfunction, polyendocrinopathy, enteropathy, X-linked) in humans. FOXP3 expression is also regulated by p53 in response to DNA damage in certain cancer cells.
