The A2MR-a2 monoclonal antibody reacts with human CD91 (also known as LRP-1 and alpha-2 macroglobulin receptor).
CD91 is a 600 kDa type I transmembrane protein composed of alpha and beta chains.

• The alpha chain binds multiple ligands.
• The beta chain contains tyrosine phosphorylation motifs interacting with adaptor and signaling proteins.

Expression: Monocytic lineage cells, dendritic cells, hepatocytes, fibroblasts, adipocytes, neurons, syncytiotrophoblast.
Functions: Mediates endocytosis, antigen cross-presentation via MHC class I, cell migration, and activation of signaling cascades.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported

• Flow cytometric analysis.

Applications Tested

• Pre-titrated and validated for flow cytometry of normal human peripheral blood cells.
• Use 5 µL (0.1 µg) per test (100 µL final volume).
• Suitable for 10⁵–10⁸ cells/test.

Usage Notes

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
• Add 5 µL of CellBlox Plus per sample prior to combining with cells.

Target Information

The encoded protein (CD91/LRP1) is an endocytic receptor involved in:

• Intracellular signaling
• Lipid homeostasis
• Clearance of apoptotic cells
• A2M-mediated clearance of amyloid precursor protein and β-amyloid

Expression decreases with age and is reduced in Alzheimer’s disease brain tissue.
