Target Information
Lipolytic enzymes are essential for mobilizing fatty acids from triglyceride stores in adipose tissue. Dysfunctional lipolysis affects energy homeostasis and may contribute to obesity and insulin resistance. Hormone-sensitive lipase (HSL) was previously known as the sole triglyceride-hydrolyzing enzyme in mammalian adipose tissue. However, adipose triglyceride lipase (ATGL) is now recognized as catalyzing the initial step in triglyceride hydrolysis. ATGL is highly expressed in adipose tissue of mice and humans, exhibits high substrate specificity for triacylglycerol, and associates with lipid droplets. Inhibition of ATGL significantly reduces total adipose acyl-hydrolase activity, indicating that ATGL and HSL coordinate triglyceride catabolism in mammalian adipose tissue.
