Target Information
Parkinson’s disease is the second most common neurodegenerative disorder after Alzheimer’s. About 1% of people over 65 and 3% over 75 are affected. Mutations in Park2 are the most common genetic cause identified to date. Park2 may play a role in the ubiquitin-mediated proteolytic pathway, and mutations can lead to autosomal recessive juvenile parkinsonism. Alternative splicing produces three known isoforms of undetermined function. Panneuronal expression of Parkin substrate Pael-R leads to age-dependent degeneration of dopaminergic neurons in Drosophila, while coexpression of Parkin degrades Pael-R and suppresses its toxicity.
