Parkinson’s disease is the second most common neurodegenerative disorder after Alzheimer’s. Approximately 1% of people over 65 and 3% over 75 are affected. Mutations in the Park2 gene are among the most common causes of Parkinson’s disease. Park2 may function in the ubiquitin-mediated proteolytic pathway. Mutations are associated with autosomal recessive juvenile parkinsonism. Alternative splicing yields three isoforms with undetermined functions. Panneuronal expression of Parkin substrate Pael-R induces age-dependent degeneration of Drosophila dopaminergic neurons; coexpression of Parkin degrades Pael-R and suppresses its toxicity.
