Description:
The monoclonal antibody CXCR3-173 recognizes mouse CD183 (CXCR3), a seven-transmembrane G-protein-linked chemokine receptor binding CXCL9 (Mig), CXCL10 (IP-10), and CXCL11 (ITAC). CD183 plays a key role in CD4 T cell responses to grafts, with CXCR3 knockout mice showing impaired allograft rejection. Expression is observed on NK cells, subsets of T lymphocytes, Tregs, and preferentially on Th1-polarized cells.
CXCR3-173 affects chemotaxis in response to ligand; ligand presence eliminates staining with the antibody. In vivo, antibody addition delays cardiac and pancreatic islet allograft rejection.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes; ≤0.5 µg/test recommended. A test is defined as the amount of antibody that stains cells in 100 µL volume (10⁵–10⁸ cells/test). Optimal titration is recommended.
Storage and Handling: Use in sterile environment.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information
This gene encodes a G protein-coupled receptor selective for chemokines IP10, Mig, and I-TAC. These ligands belong to the CXC chemokine subfamily, characterized by a single amino acid between the first two conserved cysteines. Ligand binding induces leukocyte trafficking responses such as integrin activation, cytoskeletal rearrangement, and chemotaxis.
Inhibition by Bordetella pertussis toxin suggests coupling with Gi-type G proteins. Signal transduction likely involves enzymes common to other chemokine receptor pathways. Receptor internalization and recycling regulate response duration and reactivation. Expression is prominent in effector/memory T cells and inflamed tissues, indicating a role in inflammatory cell recruitment.
This receptor is a potential therapeutic target for small-molecule antagonists in inflammatory disease treatment. Multiple transcript variants exist.
