Product Specific Information

This P84 monoclonal antibody reacts with mouse CD172a (SIRPα), a cell surface glycoprotein with three Ig-like extracellular domains and two cytoplasmic ITIM motifs. These motifs recruit SHP-1 and SHP-2 phosphatases. CD172a is expressed on monocytes, macrophages, and dendritic cells, but not on T or B lymphocytes. It is also expressed in neurons and other CNS tissues. The ligand for CD172a is CD47, and their interaction is involved in dendritic cell-mediated T cell activation, neutrophil migration, and phagocytosis.

This antibody has reported neutralizing activity.

Applications Reported: Flow cytometric analysis and neutralization assays.
Applications Tested: Tested by flow cytometric analysis of mouse bone marrow cells. Recommended ≤0.25 µg/test in 100 µL final volume. Cell number may vary (10⁵–10⁸ cells/test). Titrate for optimal performance.

Target Information

SIRPα (CD172a) is a receptor-type transmembrane glycoprotein expressed on myeloid-origin cells such as granulocytes, dendritic cells, macrophages, mast cells, and hematopoietic stem cells. It acts as a substrate for activated tyrosine kinases (EGFR, PDGFR, Src, insulin receptor) and negatively regulates receptor tyrosine kinase signaling pathways. Binding to CD47 induces ITIM phosphorylation, recruiting SHP-1 and SHP-2, and modulates macrophage phagocytosis and T cell/DC activation. SIRPα plays roles in various physiological and pathological processes, including cancer cell immune evasion.
