Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin family that plays a critical role in T cell signal transduction, differentiation, and activation. CD8 is mainly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, functioning as a co-receptor with the T cell receptor (TCR) for MHC class I molecules. In contrast, CD4 serves as a co-receptor for MHC class II molecules. CD8α is essential for binding to MHC-I and is also expressed as a homodimer on subsets of T cells, NK cells, monocytes, and dendritic cells. Engagement of MHC-I/peptide complexes by antigen-presenting cells triggers recruitment of Lck kinase, leading to cytokine production, cell motility, and activation of cytotoxic T lymphocytes (CTLs). Activated CTLs are essential for the clearance of pathogens and tumor cells. Differentiation of naïve CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
