Target Information
Circadian rhythmicity is a fundamental property across diverse organisms including animals, plants, and fungi. Regulation of endogenous biological clocks occurs via a protein-mediated autoregulatory feedback loop. In mammals, members of the basic helix-loop-helix (bHLH) PAS (PER-ARNT-SIM) transcription factor family are key regulators of circadian oscillations. CLOCK-induced genes are transactivated through E-box enhancers (CACGTG) located upstream of target genes. CLOCK-ARNT3 heterodimers bind to these regulatory elements to stimulate transcription. The PER protein, together with the mammalian timeless gene (TIM) product, negatively regulates its own transcription by inhibiting the CLOCK-BMAL1 transactivation complex.
