Target Information

Inositol 1,4,5-trisphosphate (IP3) is a second messenger for various growth factors, hormones, and neurotransmitters. Upon binding to IP3 receptor (IP3R), IP3 triggers the release of intracellular calcium from luminal stores into the cytosol. The functional IP3R is a homo- or heterotetramer composed of ~240 kDa glycoprotein subunits. IP3R is structurally and functionally related to the ryanodine receptor, another intracellular calcium-release channel. Both receptors can associate with the immunophilin FKBP12 protein, influencing calcium signaling. Phosphorylation of IP3R by PKC enhances IP3-mediated calcium release, while dephosphorylation by calcineurin reduces it, leading to calcium oscillations. Mammalian IP3R subunits arise from three distinct genes (IP3R-I, IP3R-II, IP3R-III) that are differentially expressed across tissues, including heart, liver, kidney, ovary, cerebellum, brain, pancreas, and gastrointestinal tract.
