Product Specific Information

  • Predicted to react with mouse based on sequence homology.
  • Heat-mediated antigen retrieval using sodium citrate buffer (pH 6.0) is recommended for paraffin section staining.
  • Suggested positive control: human breast carcinoma.
  • Detects a band at ~23 kDa in K562 cell lysate (Western blot).
  • Recognizes the p53-upregulated modulator of apoptosis (PUMA), also known as Bcl-2 binding component 3, JFY-1, or Bcl-2.

Target Information

PUMA is a pro-apoptotic Bcl-2 family member, along with Bax and Noxa, which are transcriptional targets of p53.
The PUMA gene encodes two BH3 domain-containing proteins, PUMA-a and PUMA-b.
PUMA proteins bind Bcl-2, localize to mitochondria, and promote cytochrome c release and apoptosis in response to p53 activation.
Antisense inhibition of PUMA expression reduces p53-mediated apoptosis, indicating its role in the cytochrome c/Apaf-1-dependent pathway.
