Target Information
ICAD (Inhibitor of Caspase-3-Activated DNase) is a substrate controlling nuclear apoptosis. It exists in two isoforms:
- ICAD-L/DFF45 (45 kDa) – functional isoform
- ICAD-S/DFF35 (35 kDa) – non-functional isoform
Both isoforms bind and inhibit CAD, but only ICAD-L is functional. Upon caspase-3 activation, ICAD is cleaved at residues 117 and 224, releasing CAD to execute apoptotic DNA fragmentation. In non-apoptotic cells, CAD remains complexed with ICAD. ICAD also acts as a CAD-specific folding chaperone.
Recent studies suggest ICAD may be a therapeutic target for restoring normal apoptotic pathways in colon and brain cancers. Two alternatively spliced transcript variants encode distinct isoforms of ICAD.
