The antibody was affinity-purified from rabbit antiserum using an epitope-specific immunogen. The purity is greater than 95% as determined by SDS-PAGE.
Target Information
RAG2 (V(D)J recombination-activating protein 2) is a core component of the RAG complex, a multiprotein complex mediating DNA cleavage during V(D)J recombination. This process assembles a diverse repertoire of immunoglobulin and T-cell receptor genes in developing B and T lymphocytes through rearrangement of V (variable), D (diversity), and J (joining) gene segments.
DNA cleavage by the RAG complex occurs in two steps:
- A nick is introduced in the top strand upstream of the heptamer, generating a 3′-hydroxyl group that attacks the opposite strand, creating four DNA ends.
- Hairpin coding ends and blunt, 5′-phosphorylated ends are generated.
Chromatin structure plays an essential role in V(D)J recombination. Histone H3 trimethylation at Lys-4 (H3K4me3) stimulates both nicking and hairpin formation. DNA breaks on one immunoglobulin allele induce ATM-dependent repositioning of the other allele to pericentromeric heterochromatin, preventing recombination of the second allele.
RAG2 is not the catalytic component but is required for all known catalytic activities mediated by RAG1. Mutations in the RAG2 gene can cause combined cellular and humoral immune defects, including granulomas, severe combined immunodeficiency (T-cell-negative/B-cell-negative/NK-cell-positive), and Omenn syndrome.
