Target Information

Progression through the cell cycle requires activation of cyclin-dependent kinases (Cdks). Cdk2, a key enzyme for cell cycle initiation, is inactive alone and requires cyclin binding and phosphorylation at Thr160 for activation. The enzyme responsible for this activation, Cdk-activating kinase (CAK), consists of a catalytic subunit, a regulatory subunit, and an additional subunit of unknown function. The regulatory subunit, cyclin H, activates Cdk7. Mat1, a protein associated with the cyclin H–Cdk7 complex, is essential for forming an active Cdk–cyclin complex and exhibits kinase activity toward Cdk activation domains and RNA polymerase II. Mat1 represents an assembly factor crucial for active Cdk–cyclin complex formation.
