Target Information
Diacylglycerol (DAG) acts as an intracellular activator of protein kinase C (PKC) and guanine nucleotide exchange factors, influencing numerous cell signaling cascades. Intracellular DAG levels are regulated by diacylglycerol kinases (DGKs), which phosphorylate DAG to phosphatidic acid.
DGKs are classified into several types:
- Type I: DGK-α (80 kDa), -β (90 kDa), -γ (90 kDa) – contain calcium-binding EF-hand motifs
- Type II: DGK-δ (130 kDa), DGK-ι (130 kDa) – contain N-terminal pleckstrin homology (PH) domains
- Type III: DGK-ε (64 kDa) – lacks identifiable regulatory domains
- Type IV: DGK-ζ (104 kDa), DGK-ι (130 kDa) – possess C-terminal ankyrin repeats
- Type V: DGK-θ (110 kDa) – contains three cysteine-rich domains and a PH domain
