Target Information
Protein p21 is a tumor suppressor protein that inhibits Cyclin-dependent kinases (CDK) and is transcriptionally activated by p53.
It plays a key role in cellular response to genotoxic stress and DNA damage repair.
In the nucleus, p21 binds and inhibits Cdk1 and Cdk2, blocking cell cycle progression from G1 to S phase or G2 to mitosis.
In the cytoplasm, p21 exhibits anti-apoptotic effects by inhibiting caspase 3, ASK1, and JNK.
It has dual roles in carcinogenesis, functioning both as a tumor suppressor and as an oncogene depending on context.
