Product Specific Information

This L161 monoclonal antibody detects CD1c (BDCA-1), a glycoprotein noncovalently linked to beta-2 microglobulin on thymocytes and antigen-presenting cells such as dendritic and Langerhans cells. It is also expressed on certain circulating and marginal zone B cells, as well as in lymph nodes and germinal centers. CD1c is involved in presenting lipid antigens (e.g., microbial fatty acids) to effector T cells during the adaptive immune response. Alternative splicing yields three isoforms: soluble, membrane, and cytoplasmic/soluble.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Recommended use at 5 µL (0.06 µg) per test in a final volume of 100 µL.

Note:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
  • Add 5 µL per sample before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

The CD1 family comprises transmembrane glycoproteins structurally related to MHC proteins, forming heterodimers with beta-2-microglobulin. These proteins present lipid and glycolipid antigens to T cells. Human CD1 genes are clustered on chromosome 1 and differ in localization and ligand specificity. CD1c localizes to endosomes and lysosomes via a tyrosine-based motif and requires vesicular acidification for lipid antigen binding.
