SMADs are members of the MAD-related family of intracellular proteins essential in the signaling pathways of serine/threonine kinase receptors within the transforming growth factor beta (TGF-β) superfamily.
SMADs are categorized into:
- R-SMADs: SMAD1, 2, 5, 8, and 9
- Co-SMAD: SMAD4
- I-SMADs: SMAD6 and 7
SMAD1, 5, 8, and 9 share high homology, and antibodies may recognize sequences common to these proteins. SMAD8 and SMAD9 are often used interchangeably in literature.
