Target Information

Caspase-activated DNase (CAD) cleaves chromosomal DNA during apoptosis. Apoptosis is a programmed cell death process that removes unwanted or damaged cells during mammalian development. It can be triggered by various stimuli such as tumor necrosis factor family death factors, UV irradiation, anticancer drugs, or survival factor deprivation.

During apoptosis, cells and nuclei shrink and fragment, and chromosomal DNA is degraded into nucleosomal units. CAD, activated by caspases, forms a complex with its inhibitor ICAD. ICAD assists in proper folding of CAD and inhibits its DNase activity. When caspases are activated, ICAD is cleaved, releasing CAD to enter the nucleus and degrade DNA.

Apoptosis regulation involves a balance between pro- and anti-apoptotic proteins of the Bcl-2 family, heat shock proteins, and inhibitors of apoptosis proteins (IAPs). Fourteen caspase family members have been identified, seven of which mediate apoptosis.
