Product Specific Information
The monoclonal antibody eBio3A3-N2 reacts with human IL-21, a potent stimulator of NK and cytolytic T cells. IL-21 is a 131 amino acid protein closely related to IL-2 and IL-15, expressed in activated CD4+ cells. It stimulates proliferation of B cells via CD40 antigen crosslinking, promotes bone marrow progenitor cell proliferation, and induces NK-cell marker CD56 expression in presence of IL-15.

Used for ELISA capture in IL-21 analysis.
Applications Tested:
Validated as capture antibody in sandwich ELISA for human IL-21 detection, in combination with biotin-labeled eBio2B2-G20 (Product #13-7218-81) and recombinant human IL-21 standard (Product #39-8219-65).
Recommended ELISA capture concentration: 1–4 µg/mL.
Include standard curve (4000 pg/mL – 60 pg/mL) in each plate.
Target Information
IL-21 is a 17 kDa immunomodulatory cytokine mainly produced by NKT, Th17, and TFH cells.
In TFH cells, IL-21 triggers autocrine signaling via IL-21R and STAT3, activating transcription factor Bcl6. It is critical for TFH cell development and function, promoting B cell differentiation into plasma and memory cells, stimulating IgG production, and inducing apoptosis in naive B cells.
In Th17 cells, IL-21 regulates IL-23R expression through STAT3, IRF4, and RORγt, promoting maturation and maintenance under IL-23 stimulation. It also downregulates Foxp3.
IL-21-deficient mice show protection against chemically induced colitis, while elevated IL-21 levels are observed in colitis models.
