Product Specific Information

This 4LURPIE monoclonal antibody recognizes human and mouse STAT4 phosphorylated on tyrosine 693 (Y693). STAT proteins are activated by cytokine receptor binding and phosphorylation by JAK family kinases, leading to nuclear translocation and gene regulation. STAT4 is activated by IL-12 or type I IFNs and is essential for Th1 differentiation.

In mice, IFNα induces transient and weak STAT4 phosphorylation, whereas in humans, IFNα promotes Th1 development and IFNγ production. Specificity was confirmed by ELISA and flow cytometry.

  • Intracellular staining followed by flow cytometric analysis.

Applications Tested

  • Pre-titrated and tested by intracellular staining of stimulated human peripheral blood cells.
    • Recommended usage: 5 µL (0.25 µg) per test (100 µL final volume).
    • Cell number range: 10⁵–10⁸ cells/test.

Target Information

STAT4 shares sequence similarity with STAT1 (52%) and STAT3 (47%) and is phosphorylated by Jak1 or Jak2. It forms homo- and heterodimers, binds the IFN-γ activated site (GAS), and requires serine phosphorylation for maximal activity. STAT4 expression is limited to thymus, spleen, and testis.

Activated by IL-12, STAT4 mediates T-cell independent IFN-γ induction, critical for defense against intracellular pathogens. STAT4-deficient mice show impaired Th1 differentiation and enhanced Th2 development. In viral infection, IFN-α/β activation of STAT4 is essential for IFN-γ production.
