Description:
The dG9 antibody clone reacts with human perforin (pore-forming protein, pfp). Perforin is a cytolytic mediator found in cytotoxic T lymphocytes (CTL) and natural killer (NK) cells, playing a critical role in immune defense against tumors and viral infections.

Applications Reported:
Reported for intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-titrated and validated for intracellular staining and flow cytometric analysis of normal human peripheral blood cells.
Recommended use: 5 µL (0.06 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Target Information

Perforin is a cytolytic protein stored in cytoplasmic granules of cytotoxic lymphocytes. It plays a key role in immune defense against tumors and viral infections by forming transmembrane pores in target cells.
It consists of 555 amino acids with a 21 amino acid signal peptide and has a molecular weight of 70–75 kDa. Perforin shares structural similarity with complement component C9. Expression is restricted to killer cell lines, not helper T cells or other tumor cells.
Mutations in the perforin gene can cause familial hemophagocytic lymphohistiocytosis type 2 (HPLH2), a rare autosomal recessive disorder. Multiple transcript variants exist due to alternative splicing.
