• For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

MHC class II molecules are transmembrane glycoproteins composed of an alpha chain (36 kDa) and a beta chain (27 kDa). They are primarily expressed on antigen-presenting cells such as B lymphocytes, monocytes, macrophages, thymic epithelial cells, and activated T lymphocytes.
Three loci—DR, DQ, and DP—encode the major expressed products of the human class II region.
Human MHC class II molecules bind intracellularly processed peptides and present them to T-helper cells, playing a critical role in the initiation of immune responses.

HLA and MHC antibodies are important tools in immunopeptidomics, enabling the identification and characterization of neoantigens through high-performance liquid chromatography coupled with tandem mass spectrometry.
