Target Information
Beta-catenin은 세포 간 부착 구조(adherens junction, AJ)의 구성 요소로 처음 확인된 단백질입니다. AJs는 세포 성장과 세포 간 부착을 조절하여 상피 세포층의 생성과 유지에 필수적입니다. Beta-catenin은 E-cadherin의 세포질 도메인과 상호작용하여 E-cadherin을 alpha-catenin에 연결하며, 이는 E-cadherin 복합체를 세포골격(actin cytoskeleton)에 고정시키는 역할을 합니다. 또한 Beta-catenin은 adenomatous polyposis coli 단백질 및 미세소관을 포함하는 세포골격 복합체와 결합하며, tyrosine kinase, phosphatase, Wnt/Wingless 등 다양한 신호전달 경로와 상호작용합니다.
이러한 상호작용은 형태형성과 세포 증식 조절에 관여하며, Beta-catenin은 모낭세포, 기저세포, 외부 및 내부 뿌리초 세포 등에서 발현됩니다. 병리학적으로는 pilomatrixoma, medulloblastoma, colorectal cancer, ovarian cancer 등 여러 종양 발생과 관련이 있습니다. 핵 내에서는 Lef/Tcf 전사인자와 결합하여 cyclin D 및 c-myc 유전자의 전사를 촉진함으로써 세포 증식을 유도합니다.
