Target Information
Beta-catenin은 세포-세포 접착 구조의 구성 요소로 처음 확인된 adherens junction(AJ) 단백질입니다. AJ는 세포 성장과 접착을 조절하여 상피세포층의 형성과 유지에 필수적입니다. Beta-catenin은 E-cadherin의 세포질 도메인과 상호작용하여 E-cadherin을 alpha-catenin에 연결하며, 이는 E-cadherin 복합체를 피질 액틴 세포골격에 고정시킵니다. 또한 Beta-catenin은 adenomatous polyposis coli 단백질 및 미세소관과 결합하는 세포골격 복합체와 상호작용하며, tyrosine kinase, phosphatase 및 Wnt/Wingless를 포함한 여러 신호 전달 경로와 관련됩니다.
이러한 복합체 및 신호 경로 간의 상호작용은 형태형성(morphogenesis)을 조절할 수 있습니다. Beta-catenin은 모낭 세포, 기저 및 주변 기질 세포, 외부 및 내부 뿌리초 세포 등 다양한 세포 유형에서 발현됩니다. 병리학적으로는 pilomatrixoma, medulloblastoma, colorectal cancer, ovarian cancer 등 다양한 종양에서 역할이 보고되었습니다. 핵 내에서 Beta-catenin은 Lef/Tcf 전사인자군을 공동활성화하여 cyclin D 및 c-myc 유전자의 발현을 촉진하며, 이는 세포 증식 및 종양 발생과 관련됩니다.
