Target Information

PTEN was identified as a tumor suppressor frequently mutated in various cancers. The PTEN gene encodes a phosphatidylinositol-3,4,5-trisphosphate 3-phosphatase containing a tensin-like domain and a catalytic domain similar to dual specificity protein tyrosine phosphatases. Unlike most tyrosine phosphatases, PTEN preferentially dephosphorylates phosphoinositide substrates. It negatively regulates intracellular phosphatidylinositol-3,4,5-trisphosphate levels and functions as a tumor suppressor by inhibiting the AKT/PKB signaling pathway. PTEN also modulates the AKT-mTOR pathway, influencing neuronal integration, dendritic development, and synapse formation during adult neurogenesis. Additionally, PTEN may act as a negative regulator of insulin signaling and glucose metabolism in adipose tissue. The nuclear monoubiquitinated form has higher apoptotic potential, while the cytoplasmic nonubiquitinated form shows reduced tumor suppressive activity.
