Target Information

PTEN was identified as a tumor suppressor frequently mutated in various cancers. The PTEN gene encodes a phosphatidylinositol-3,4,5-trisphosphate 3-phosphatase containing a tensin-like and a catalytic domain similar to dual-specificity protein tyrosine phosphatases. Unlike most phosphatases, PTEN preferentially dephosphorylates phosphoinositide substrates and negatively regulates intracellular PIP3 levels, thereby inhibiting the AKT/PKB signaling pathway.
PTEN is a key modulator in the AKT-mTOR signaling pathway, influencing neuronal integration, positioning, dendritic development, and synapse formation during adult neurogenesis. It may also act as a negative regulator of insulin signaling and glucose metabolism in adipose tissue. The nuclear monoubiquitinated form exhibits greater apoptotic potential, while the cytoplasmic nonubiquitinated form shows reduced tumor suppressive ability.
