Description:
The eBioCB-CALLA monoclonal antibody recognizes human CD10 (CALLA, NEP, enkephalinase, Neprilysin), a 100 kDa type II cell surface glycoprotein originally identified for its expression on most acute lymphoblastic leukemias (ALL). CD10 is a Zn²⁺-dependent metallo-peptidase that regulates chemotactic and inflammatory processes involving neutrophils and is expressed in B cells, enterocytes, and renal tubules. Partial blocking occurs between eBioCB-CALLA and MEM-78 antibodies, indicating recognition of similar epitopes.

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use at 5 µL (0.125 µg) per test. A test is defined as the amount of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically (10⁵–10⁸ cells/test).

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be determined empirically.

488–561 nm / 775 nm (Blue, Green, Yellow-Green Laser)

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

CD10 (Common Acute Lymphocytic Leukemia Antigen, CALLA) is a cell surface enzyme with neutral metalloendopeptidase activity that inactivates various biologically active peptides. It is expressed on lymphoblastic, Burkitt’s, and follicular germinal center lymphomas, immature B cells, and on cells from chronic myelocytic leukemia (CML) patients. CD10 is also found on breast myoepithelial cells, bile canaliculi, fibroblasts, and highly expressed on kidney and gut epithelial cells. It cleaves peptides at the amino side of hydrophobic residues and inactivates several peptide hormones including glucagon, enkephalins, substance P, neurotensin, oxytocin, and bradykinin. The CD10 gene produces multiple mRNA transcripts through alternative splicing.
