Product Specific Information
Description:
The RPA-T8 monoclonal antibody reacts with the human CD8a molecule (~32–34 kDa), expressed as CD8α/β heterodimer or CD8α/α homodimer. CD8a is found on thymocytes, mature T cells, and NK cells. It binds to MHC class I and associates with p56lck kinase, playing a key role in T-cell development and activation.
Applications Reported:
Suitable for flow cytometric analysis.
Applications Tested:
Pre-titrated for flow cytometry of normal human peripheral blood cells. Use at 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
Light Sensitivity:
This tandem dye is photo-sensitive; protect vial and samples from light.
Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-822-49) or 1-step Fix/Lyse Solution (Product #00-5333-54).
Excitation / Emission / Laser:
488–561 nm excitation; 775 nm emission; compatible with Blue, Green, and Yellow-Green lasers.
Filtration:
0.2 µm post-manufacturing filtered.
Target Information
Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It functions as a co-receptor with TCR for MHC class I molecules and is essential for T-cell activation and differentiation. CD8 is expressed on T cells, NK cells, monocytes, and dendritic cells as CD8α/β heterodimers or CD8α/α homodimers. MHC-I/peptide ligation triggers Lck kinase activation, leading to CTL responses critical for pathogen and tumor clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
