Product Specific Information

This antibody neutralizes natural and recombinant rat IFN-γ. Cross-reactivity to human IFN-γ is less than 0.01% of the homologous combination.
Biological Activity: >2.5 × 10⁴ neutralizing units/mg, where one neutralizing unit is defined as the amount of antibody sufficient for neutralizing one unit recombinant rat IFN-γ (~150 pg) in an antiviral activity bioassay.

For immunohistochemistry applications, it is recommended that the reconstituted antibody be dialyzed against PBS to remove trehalose.

Storage and Reconstitution:

• Store the lyophilized antibody at 2–8°C.
• Reconstitute using 0.5 mL sterile distilled water.
• After reconstitution, store at 2–8°C for up to several weeks.
• For maximal stability, apportion into aliquots and store at -20°C, avoiding repeated freeze/thaw cycles.
• 0.02% sodium azide may be added to prevent bacterial growth.

Target Information

IFN gamma (Interferon gamma, Type II interferon) is a macrophage activation factor and immune interferon produced primarily by T-lymphocytes and natural killer cells in response to antigens, mitogens, and other cytokines.
It is a dimeric glycoprotein composed of two 146 amino acid subunits (~45 kDa as a homodimer).
On SDS-PAGE, IFN gamma appears as 25, 20, and minor 15.5 kDa bands due to differential glycosylation.

• Antiviral and tumor antiproliferative activity
• Induction of class I and II MHC
• Macrophage activation
• Enhanced immunoglobulin secretion by B lymphocytes
• Cytokine regulation and synergy with other cytokines

Activation occurs via IFN gamma receptor I and II, triggering the JAK-STAT pathway.
Human IFN gamma does not show cross-reactivity with mouse IFN gamma but shares ~40% sequence homology.
IFN gamma expression is upregulated by IL2, FGF basic, EGF, and downregulated by vitamin D3 or DMN.
Gene mutations are associated with aplastic anemia.
