Product Specific Information

This HMER5DS antibody recognizes human MerTK (Mer or c-mer), a 170–210 kDa member of the TAM family of tyrosine kinase receptors that includes Axl and Tyro3.
MerTK is expressed on a subset of anti-inflammatory macrophages and is involved in the removal of apoptotic cells via ligands Protein S and Gas6.
Upon ligand binding, MerTK undergoes autophosphorylation, activating PI3K and Akt pathways, leading to phagocytosis and inhibition of TLR-induced cytokine production.
Mer may also act as an entry receptor for filoviruses. Deficiency causes autoimmunity and inflammation. MerTK expression is often upregulated in malignant cells and may contribute to immune evasion.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated for human monocyte-derived macrophages at 5 µL (0.06 µg)/test.
A test is defined as the amount (µg) of antibody staining a cell sample (100 µL final volume).
Cell number: 10⁵–10⁸ cells/test (empirical).
Excitation: 488–561 nm; Emission: 578 nm; Laser: Blue, Green, Yellow-Green.
Filtration: 0.2 µm post-manufacturing filtered.


Target Information

MERTK (cMER) is a tyrosine kinase proto-oncogene involved in retinal pigment epithelium (RPE) phagocytosis and other processes including cell survival, migration, differentiation, and efferocytosis.
Ligand binding induces autophosphorylation and downstream signaling via GRB2 or PLCG2, activating MAPK, FAK/PTK2, and RAC1 pathways.
MERTK plays roles in macrophage clearance of apoptotic cells, platelet aggregation, cytoskeletal reorganization, and inhibition of TLR-mediated innate immune responses through STAT1 activation and SOCS1/3 induction.
Functions as a regulator of rod outer segment fragment phagocytosis in RPE.
