Protein kinases are enzymes that transfer a phosphate group from a phosphate donor, generally the γ phosphate of ATP, onto an acceptor amino acid in a substrate protein. Through this mechanism, they mediate most signal transduction in eukaryotic cells, regulating metabolism, transcription, cell cycle, cytoskeletal rearrangement, cell movement, apoptosis, and differentiation.

It is classified into eight major groups based on catalytic domain sequences of tyrosine (PTK) or serine/threonine (STK) kinases. The tyrosine-like kinase (TLK) group includes MLK, LISK, IRAK, Raf, RIPK, and STRK families.
