Description:
The MIH24 monoclonal antibody reacts with human TRANCE (TNF-related activation-induced cytokine receptor), also known as RANKL (receptor activator of NF-kappaB ligand) and OPGL (Osteoprotegerin Ligand). TRANCE is a type II membrane protein (~35 kDa) with an extracellular domain closely related to TRAIL, FasL, and TNF. Its expression is upregulated by TCR stimulation, and TRANCE-deficient mice lack osteoclasts. TRANCE is involved in T-dendritic and T-B cell interactions and is critical for osteoclast differentiation. The TRANCE-RANK interaction regulates expression of the anti-apoptotic molecule Bcl-xL.

Applications Tested:
Tested by flow cytometric analysis of human RANKL transfected cells. Recommended ≤1 µg/test (in 100 µL final volume). Cell number should be empirically determined (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal performance.

Target Information
This gene encodes a member of the TNF cytokine family, a ligand for osteoprotegerin, and a key factor for osteoclast differentiation and activation. It acts as a dendritic cell survival factor and regulates T cell-dependent immune responses. T cell activation induces expression of this gene, promoting osteoclastogenesis and bone loss. The protein activates antiapoptotic kinase AKT/PKB via a complex involving SRC and TRAF6, suggesting a role in apoptosis regulation. Gene disruption in mice causes osteopetrosis, lack of osteoclasts, and defects in T/B lymphocyte differentiation and mammary gland development. Two alternatively spliced transcript variants are known.
