Product Specific Information

The PK136 monoclonal antibody reacts with mouse NK1.1, an antigen expressed by natural killer (NK) cells and a subset of T cells in NK1.1 mouse strains including C57BL and NZB.
Common laboratory mouse strains such as BALB/c, SJL, AKR, CBA, C3H, and A do not express NK1.1. For these strains, the DX5 monoclonal antibody (Product # 14-5971) should be used.

Simultaneous staining of C57BL/6 spleen cells with PK136 and DX5 shows coexpression of both markers by most cells and presence of DX5⁺PK136⁻ and DX5⁻PK136⁺ populations.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on C57Bl/6 splenocytes at ≤1 µg/test (100 µL final volume). Cell number should be empirically determined (10⁵–10⁸ cells/test).
Recommendation: Titrate antibody for optimal performance.

eFluor® 450 is an alternative to Pacific Blue®, excited by a 405 nm violet laser and emitting at 445 nm. Ensure instrument compatibility.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

NK1.1 (Killer cell lectin-like receptor subfamily B, member 1; KLRB1; NKR-P1A; CD161) is a marker for natural killer (NK) cells, which mediate cytotoxicity and secrete cytokines upon immune stimulation.
KLRB1 protein belongs to the C-type lectin superfamily and contains extracellular, transmembrane, and cytoplasmic domains.
NKR-P1A (CD161) recognizes Lectin Like Transcript-1 (LLT1) as its functional ligand and is classified as a type II membrane protein with an external C-terminus.
