Product Specific Information

The MRa4-1 monoclonal antibody reacts with CD49d, also known as integrin alpha 4 subunit. This 150-kDa surface protein is expressed by thymocytes, peripheral lymphocytes, monocytes, and eosinophils, forming complexes with integrin beta1 or beta7.
Outside the immune system, CD49d is expressed on mast cells, neural crest cells, retinal cells, and dorsal root ganglion neurons. Integrins are involved in lymphocyte homing to Peyer’s patches and mesenteric lymph nodes, as well as neural development and regeneration.
Ligands for the integrin α4β1 complex include VCAM-1 (CD106) and fibronectin.
The MRa4-1 antibody can inhibit fibronectin binding and lymphocyte migration.

Target Information

Integrin alpha 4 (CD49d, ITGA4) is a member of the integrin alpha chain family. Integrins are heterodimeric membrane proteins composed of alpha and beta chains. The alpha4 chain associates with beta1 or beta7 chains.
Ligand-binding sites of both α4β1 and α4β7 are located on the alpha4 chain. Known ligands include Madcam, VCAM, and fibronectin. Madcam is the principal ligand for α4β7.
HIV-1 envelope can mimic Madcam by binding and signaling through integrin α4β7, the gut mucosal homing receptor for peripheral T cells.
