Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. Upon activation, p53 induces transcription of genes such as p21waf1, regulating G1 or G2/M arrest.

p53 consists of an N-terminal transactivation domain, a central DNA-binding domain, an oligomerisation domain, and a C-terminal regulatory domain. Multiple phosphorylation sites exist on p53; phosphorylation at Ser15 is critical for activation and stabilization. Phosphorylation at Ser392 is associated with tumor formation.

p53 plays a vital role in preventing proliferation of damaged cells, acting as an anticancer agent. It is also linked to aging, oxidative stress, and cognitive functions such as learning and memory. While expressed at low levels in normal cells, p53 is highly expressed in transformed cell lines, contributing to malignancy. Mutations in TP53 can lead to loss of tumor suppressor activity and are found in various cancers and hereditary cancer syndromes such as Li-Fraumeni syndrome.
