Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. p53 upregulation activates transcription of genes such as p21waf1, leading to G1 or G2/M arrest. Structurally, p53 includes an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain.

Phosphorylation at Ser15 is critical for p53 activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 plays a key role in preventing proliferation of damaged cells and acts as an anticancer factor. Elevated p53 levels have been linked to aging, oxidative stress, and cognitive deficits related to long-term potentiation. In normal cells, p53 is expressed at low levels, but it is upregulated in transformed cells, contributing to malignancy. Mutations in TP53 can lead to loss of tumor suppressor activity and are implicated in various cancers, including germline mutations in Li-Fraumeni syndrome.
