Product Specific Information

Description:
The eBioMC-480 (MC-480) antibody reacts with stage-specific embryonic antigen-1 (SSEA-1), a carbohydrate epitope expressed on early mouse embryos, murine embryonal carcinoma (EC) cells, embryonic stem (ES) cells, and murine/human germ cells (EG). No immunoreactivity is seen with undifferentiated human EC and ES cells. Differentiation of human EC increases SSEA-1 expression, while in mouse cells it decreases. SSEA-1 is also expressed on mature human granulocytes (CD15) and some monocytes, and is associated with cell adhesion, migration, and differentiation.

Applications Reported:
Reported for use in flow cytometry, immunoprecipitation, and immunohistochemistry.

Applications Tested:
Tested by flow cytometric analysis of the F9 cell line. Recommended ≤0.5 µg/test in 100 µL volume (10⁵–10⁸ cells/test). Titrate for optimal performance.

Target Information

CD15 (Lewis X, Le(x); SSEA-1) is a trisaccharide determinant (3-fucosyl-N-acetyllactosamine) expressed on glycolipids, glycoproteins, and proteoglycans of various cell types including granulocytes, mast cells, monocytes, macrophages, gastric mucosa cells, nervous system cells, and tumor cells. Variants include sialyl-Lewis x and sulphated Lewis x.
Cells with high Le(x) expression show calcium-dependent self-aggregation, relevant in embryo compaction and teratocarcinoma cell aggregation.
Sialyl-Le(x) and sialyl-Le(a) act as selectin ligands. CD15 expression is widely used in the diagnosis of Hodgkin’s disease.
