Product Specific Information
The eBioclone 7 monoclonal antibody was generated against a mouse c-Met-Ig fusion protein and reacts with mouse c-Met-transfected cells. Mouse c-Met (HGFR) is a 170 kDa receptor tyrosine kinase (RTK) expressed by epithelial cells of the brain, kidney, liver, and other tissues. Binding of its ligand, Hepatocyte Growth Factor (HGF), triggers receptor autophosphorylation and activation of several downstream effectors including ERK-1, ERK-2, and PLCγ. Activation of the c-Met signaling pathway induces cellular responses such as motility, scattering, proliferation, survival, and angiogenesis. Mutations in human c-Met have been implicated in multiple malignancies.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse c-Met-transfected 293T cells. Recommended ≤ 1 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Titrate for optimal performance.
Purity: >90% (SDS-PAGE)
Aggregation: <10% (HPLC)
Filtration: 0.2 µm post-manufacturing filtered
Target Information
MET (c-MET) is a receptor-like tyrosine kinase linked to various human malignancies. Upon ligand activation, MET interacts with PI3-kinase subunit PIK3R1, PLCG1, SRC, GRB2, and STAT3, leading to activation of signaling cascades including RAS-ERK, PI3K-AKT, and PLCγ-PKC. MET plays critical roles in embryonic development (gastrulation, muscle and neuron development, angiogenesis, kidney formation) and adult processes (wound healing, organ regeneration, tissue remodeling). Aberrant MET signaling is associated with cancers such as gastric, renal, and breast cancer, making it a key target for cancer therapeutics and diagnostics.
