Product Specific Information

The HI100 monoclonal antibody reacts with human CD45RA, a 220 kDa molecule expressed by subpopulations of CD4+ and CD8+ peripheral T lymphocytes and B cells. CD45RA+ T cell populations are mainly naive/virgin, allowing HI100 mAb to serve as a phenotypic marker for T cell subset discrimination.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells at 5 µL (0.8 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Required for optimal staining with NovaFluor conjugates. Add 5 µL per sample, regardless of the number of NovaFluors used. For single-color controls, use 5 µL per 100 µL of cell sample containing 10³–10⁸ cells.

NovaFluor conjugates are based on Phiton™ technology utilizing nucleic acid dye structures for engineered fluorescent signatures, minimizing spillover and spread impacts.

Target Information

CD45RA is an isoform of the CD45 complex expressed in specific lymphoid subtypes. CD45 (leukocyte common antigen) is a receptor-type protein tyrosine phosphatase found in all nucleated hematopoietic cells, comprising ~10% of lymphocyte surface proteins. It regulates Src-family kinases, lymphocyte development, and antigen signaling. Isoforms differ by extracellular domain splicing and influence membrane localization and signaling behavior.

CD45 plays roles in:

• Immunoreceptor signaling regulation
• Integrin-mediated survival signaling
• DNA fragmentation during apoptosis

Expressed on hematopoietic lineage cells except mature red cells. Dysfunction is linked to severe combined immunodeficiency and Hepatitis C.
