Cellular Localization Isoform 1: Cell membrane; Single-pass type I membrane protein; Early endosome; Cytoplasm (perinuclear region); Nucleus. Translocation to the nucleus requires endocytosis and importin beta-1/KPNB1 mediation. Also detected in VPS35-positive endosome-to-TGN retrograde vesicles.

Background This gene encodes a member of the epidermal growth factor (EGF) receptor family of receptor tyrosine kinases. The protein lacks its own ligand-binding domain but forms heterodimers with ligand-bound EGF receptor family members, stabilizing ligand binding and enhancing downstream signaling via MAPK and PI3K pathways. Allelic variations at amino acid positions 654 and 655 (isoform a) have been reported. Amplification or overexpression is associated with cancers such as breast and ovarian tumors. Alternative splicing results in several transcript variants.
