Product Specific Information

Description:
The OKT3 monoclonal antibody reacts with an epitope on the epsilon-subunit within the human CD3 complex. It has potent immunosuppressive properties in vivo and has been used effectively in the treatment of renal, heart, and liver allograft rejection. CD3 subunits (gamma, delta, epsilon) are essential for proper TCR complex assembly and expression. CD3 is expressed by thymocytes and all mature T cells. Crosslinking of TCR triggers intracellular signaling leading to activation and proliferation.

Applications Reported:
Flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.5 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Excitation / Emission:
488–561 nm / 578 nm; compatible with Blue, Green, and Yellow-Green lasers.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

The CD3 complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. It includes CD3 gamma, delta, epsilon, and zeta subunits, which are members of the immunoglobulin superfamily encoded on chromosome 11. These subunits associate with cytoplasmic signaling molecules through tyrosine-based motifs. CD3 is expressed on most T lymphocytes and thymocytes but not on B or NK cells. Defects in CD3 genes are associated with CD3 immunodeficiency.
