Product Specific Information

• This antibody does not react with rat.
• For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

HLA-DR, like other MHC class II molecules, is a transmembrane glycoprotein composed of a 36 kDa alpha chain (DRA) and a 27 kDa beta chain (DRB).
The alpha chain gene contains 5 exons:

• Exon 1 encodes the leader peptide
• Exons 2 and 3 encode the two extracellular domains
• Exon 4 encodes the transmembrane domain and cytoplasmic tail

DRA lacks polymorphisms in the peptide-binding region and acts as the sole alpha chain for DRB1, DRB3, DRB4, and DRB5. The beta chain contains the polymorphisms that determine peptide-binding specificity.
Hundreds of DRB1 alleles have been described, and typing for these polymorphisms is routinely performed for bone marrow and kidney transplantation.

HLA-DR is expressed primarily on antigen-presenting cells such as B lymphocytes, monocytes, macrophages, thymic epithelial cells, and activated T lymphocytes.
The human MHC class II region includes DR, DQ, and DP loci, which encode the major expressed products.
These molecules bind intracellularly processed peptides, present them to T-helper cells, and play a critical role in initiating immune responses.
