Product Specific Information

PA1-963 detects the proteasome 20S C2 subunit from canine, hamster, human, mouse, and rat tissues and cells.
It has been successfully used in Western blot procedures, detecting a 29 kDa protein representing the proteasome 20S C2 subunit from rat brain extract.
The immunizing peptide corresponds to amino acid residues 249–263 from the C-terminus of human proteasome 20S C2 subunit.
This sequence is 93% conserved in rat and chicken proteins and 87% conserved in mouse protein.
The immunizing peptide (Cat. # PEP-100) is available for neutralization and control experiments.


Target Information

Proteolytic degradation is essential for maintaining appropriate levels of short-lived and regulatory proteins involved in metabolism, stress response, antigen presentation, receptor modulation, cell cycle regulation, transcription, and signaling.
The ubiquitin-proteasome pathway degrades most cytosolic and nuclear proteins, while others are processed through the vacuolar pathway involving endosomes, lysosomes, and the endoplasmic reticulum.
The 26S proteasome is an ATP-dependent, multisubunit (~31), barrel-shaped complex (~2.5 MDa) consisting of a 20S core and one or two 19S regulatory subunits.
The 19S subunits recognize ubiquitinated proteins and facilitate their unfolding and translocation into the 20S core.
Defects in this pathway are associated with diseases such as cystic fibrosis, Angelman’s syndrome, and Liddle syndrome.
