Product Specific Information
The M5/114.15.2 monoclonal antibody reacts with mouse MHC class II molecules (I-A and I-E subregion-encoded glycoproteins: I-A^b, I-A^d, I-A^q, I-E^d, I-E^k, not I-A^f, I-A^k, or I-A^s). It detects determinants on B cells, monocytes, macrophages, dendritic cells, and activated T lymphocytes from mice carrying H-2^b, H-2^d, H-2^q, H-2^p, H-2^r, and H-2^u haplotypes.
Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤0.06 µg/test (100 µL final volume). Optimal titration recommended.
Super Bright 702 is a tandem dye excited by the violet laser (405 nm) and emits at 702 nm. Recommended filter: 710/50 bandpass. Ensure instrument compatibility.
When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.
Light Sensitivity: Protect vial and samples from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark.

Target Information
MHC class II molecules are transmembrane glycoproteins expressed on professional antigen-presenting cells (macrophages, dendritic cells, B cells). They bind exogenous antigens and present them to T cells. The antigen-binding groove between alpha and beta chains is open at both ends and spans 15–24 amino acids.
HLA and MHC antibodies are crucial in Immunopeptidomics for identifying and characterizing neoantigens via LC-MS/MS.
