Product Specific Information
The DTA-1 monoclonal antibody reacts with mouse GITR (Glucocorticoid-Induced TNFR family gene, TNFRSF18). In naive mice, GITR is mainly expressed by CD4⁺CD25⁺ T regulatory cells (Treg) and by CD25⁺ CD4⁺ CD8⁻ thymocytes. Stimulation of GITR with DTA-1 antibody abrogates Treg-mediated suppression and can induce tumorogenesis and organ-specific autoimmune disease.

Applications Tested: Tested on mouse splenocytes; ≤0.03 µg/test recommended (100 µL final volume per test, 10⁵–10⁸ cells/test).
Super Bright 702 is a tandem dye excited by a 405 nm violet laser and emits at 702 nm. Recommended bandpass filter: 710/50. Ensure instrument compatibility.
When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize polymer interactions.
Light Sensitivity: Protect vial and stained samples from light.
Fixation: Samples may be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information
Tumor necrosis factor receptor superfamily member 18 (TNFRSF18), also known as GITR or AITR, is encoded by the TNFRSF18 gene located at 1p36.33. It plays a key role in immunological self-tolerance mediated by CD25⁺CD4⁺ regulatory T cells and is upregulated upon T-cell activation. Knockout studies indicate involvement in CD3-driven T-cell activation and apoptosis. Three alternatively spliced transcript variants encoding distinct isoforms have been reported.
