Product Specific Information
The RMT3-23 monoclonal antibody reacts with mouse CD366 (TIM3), a Th1-specific cell surface protein expressed by both BALB/c and C57BL/6 strains.
CD366 is a type I transmembrane protein containing immunoglobulin and mucin-like domains extracellularly and a tyrosine phosphorylation motif intracellularly.
It is selectively expressed by differentiated CD4⁺ Th1 and CD8⁺ Tc1 cells, but absent on Th2 and Tc2 cells. Other hematopoietic cells do not express CD366 at the protein level.
CD366 expression is upregulated at a late stage of T cell differentiation, suggesting its role in Th1 effector function. In EAE models, CD366 is expressed on most CD4⁺ and CD8⁺ T cells in the CNS at disease onset, while peripheral expression remains low.
Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤0.5 µg/test (100 µL final volume). Optimal titration recommended.
Fluorochrome Information:
- Super Bright 702 is a tandem dye excited with the violet laser (405 nm) and emits at 702 nm.
- Recommended filter: 710/50 bandpass.
- Use Super Bright Complete Staining Buffer (Product # SB-4401) when using multiple Super Bright dyes.
- Light sensitive: Protect vial and samples from light.
- Fixation: Samples can be stored up to 3 days in the dark at 4°C using IC Fixation Buffer (Product # 00-8222) or Fix/Lyse Solution (Product # 00-5333).

Target Information
TIM3 (Hepatitis A virus cellular receptor 2, HAVCR2) is a 281 amino acid, type I Th1-specific glycoprotein expressed on terminally differentiated CD4⁺ Th1 and CD8⁺ Tc1 cells.
It consists of an IgV-like domain, a mucin-like domain, and a conserved tyrosine phosphorylation motif.
TIM3 plays roles in macrophage activation, autoimmune disease induction, and immune tolerance regulation.
Its pathological significance includes involvement in experimental autoimmune encephalomyelitis (EAE), enhancing disease severity in mice.
