Target Information
Phosphoinositide-specific phospholipase C (PLC)는 수용체 매개 신호전달의 시작에서 중요한 역할을 하며, phosphatidylinositol 4,5-bisphosphate로부터 두 가지 2차 신호전달물질인 inositol 1,4,5-triphosphate와 diacylglycerol을 생성합니다.
PLC는 다양한 이소형을 가지며, PLCβ1, PLCβ2, PLCβ3, PLCβ4, PLCγ1, PLCγ2, PLCδ1, PLCδ2 및 PLCε 등이 포함됩니다.
PLCδ는 네 가지 isoform으로 존재하며, PLCδ1은 칼슘 신호 증폭기로서 비정형 GTP 결합 단백질에 의해 활성화됩니다. 또한 PLCδ1은 transglutaminase II 매개 oxytocin receptor 및 α1B-adrenoreceptor 신호전달의 효과기 역할을 합니다.
Mouse PLCδ1은 뇌, 심장, 폐, 고환에서 높은 발현을 보이며, Alzheimer’s disease (AD) 환자의 뇌에서 비정상적으로 축적되어 질병 병리와 관련 있을 가능성이 제시되었습니다.
PLCδ2는 type II intestinal metaplasia 및 adenocarcinoma에서 현저히 발현되며, type I intestinal metaplasia에서의 발현은 양성으로 간주되지만 종양성 변형으로 발전할 수 있습니다. 따라서 PLCδ2 발현은 위암 악성 변형의 가능성 지표로 고려될 수 있습니다.
