Description:
The 24-31 monoclonal antibody reacts with human CD154, a 39 kDa transmembrane glycoprotein also known as gp39 and CD40 ligand (CD40L). CD154 is a member of the TNF superfamily and is expressed transiently by activated T cells. Through its binding to CD40 on antigen-presenting cells (B cells, monocytes/macrophages, dendritic cells), CD154 plays a crucial role in T-APC interaction. CD154-CD40 binding induces T-dependent B-cell activation and B-cell cycle entry. The 24-31 clone cross-reacts with cynomolgus monkey CD154.

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometry using stimulated normal human peripheral blood cells. Recommended use: 5 µL (0.06 µg) per test in 100 µL final volume. Suitable for 10⁵–10⁸ cells/test.

Fluorochrome Information:
PerCP-eFluor® 710 can replace PE-Cy5, PE-Cy5.5, or PerCP-Cy5.5.

• Recommended filters: 685 LP dichroic mirror and 710/40 band pass filter (695/40 acceptable)

PerCP-eFluor® 710 conjugated antibodies are stable after overnight exposure to 2% formaldehyde at 4°C. Evaluate for alternative fixation protocols as needed.

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD40 Ligand (CD40-L, CD154) is a membrane glycoprotein expressed on T cells. It stimulates B cell proliferation and immunoglobulin secretion in the presence of cytokines. CD40 Ligand also induces cytokine production and tumoricidal activity in monocytes and co-stimulates T cell proliferation with IFN-γ, TNF-α, and IL-2 production.
